Rule of thumb on when to Create a new Domain versus using OUs I don't want to steer you wrong, but I always say if you need different security requirements go with a new domain (remember things like password policies apply to the whole domain and not OU's), but if you need to delegate administrative responsibilities go with an OU.

Making a new domain is usually more based on politics than it is based on infrastructure. Rule of Thumb on creating a new site is usually if the office that your bringing up can't be connected to your main office thru LAN connectivity you want to make a site for that office. WAN speeds are hard to determine since they change often and things like utilization and saturation can effect it. Just get a good meal and a good nights sleep tonight and you will do great tomorrow.
