Windows 2000 and beyond, no. You have to reformat the drive. The defrag utility in Windows does not support 12-bit FAT partitions. I could not find any other information.
I have no idea if there are 3rd party defrag utilities that do support it.

To work around this problem, back up the volume, reformat the volume, and then restore the backup to the volume. The data should be restored contiguously
